چکیده
Abstract Background/Introduction Obesity and metabolic dysregulation are closely associated with the pathophysiology of multiple cardiovascular diseases (CVD). To date, pathophysiological mechanism(s) obesity its link systems remain largely unknown. Adipose tissue inflammation as a result excessive fat expansion in obesity, leading to increased systemic production growth factors recruitment inflammatory cells have been postulated be major factor. Secreted frizzled-related protein 5 (SFRP5) is an anti-inflammatory adipokine that linked regulation has indicated affect functions. Currently, role circulating SFRP5 levels biomarker for poorly understood, studies yielding discordant results. Purpose This study aims evaluate relationship between functions cohort patients established CVD. Methods Patients (n=262, 148 male (56.5%), age (68±11 yrs)) presenting cardiology unit investigations were recruited into study. Plasma measured via enzyme-linked immunosorbent assay (ELISA). Associations plasma levels, functions, patients' co-morbidities analysed using univariate multivariate analyses. Results significantly lower with: heart failure (HF) vs non-HF (median; (10.7 31.0; p<0.001); coronary artery disease (CAD) non-CAD; (11.0 33.8; atrial fibrillation (AF) non-AF; (11.2 23.2; p=0.001). On analyses, also positively correlated left ventricular ejection fraction (LVEF) (r=0.52, p<0.001), estimated glomerular filtration rate (eGFR) (r=0.16, p=0.02), total cholesterol triglycerides (r=0.29, p<0.001; r=0.17, p<0.01 respectively). Low high C-reactive (CRP) E/E' (r=−0.29, p<0.001, r=−0.30, HF, CAD, statin use, low LVEF, triglycerides, CRP eGFR independent age, BMI or diabetes on analysis (overall model r=0.729, SE=0.638). Conclusion Our results show independently impaired systolic diastolic These suggest may regulate regulations. Funding Acknowledgement Type funding sources: Private grant(s) and/or Sponsorship. Epigenetic inactivation of the secreted frizzled-related protein-5 (SFRP5) gene in human breast cancer is associated with unfavorable prognosis.
Disruption of the Wnt pathway is thought to be crucial in the development of human cancer. Pathway inhibitory members of the secreted frizzled-related protein (SFRP) family were found to be downregulated due to epigenetic inactivation in various malignancies. To date, only SFRP1 has been studied in human breast cancer and we questioned whether other SFRP genes may be implicated in the pathogene...
متن کاملInverse associations between serum levels of secreted frizzled-related protein-5 (SFRP5) and multiple cardiometabolic risk factors: KORA F4 study
AIMS Secreted frizzled-related protein (Sfrp)5 has beneficial effects on insulin sensitivity, inflammation and cardiovascular risk in different mouse models, but its relevance for cardiometabolic diseases in humans is controversial. We aimed to characterise associations of circulating SFRP5 with cardiometabolic risk factors and prediabetes/type 2 diabetes in a large population-based cohort. M...
متن کاملInhibition of secreted frizzled-related protein 5 improves glucose metabolism.
Elucidating the role of secreted frizzled-related protein 5 (SFRP5) in metabolism and obesity has been complicated by contradictory findings when knockout mice were used to determine metabolic phenotypes. By overexpressing SFRP5 in obese, prediabetic mice we consistently observed elevated hyperglycemia and glucose intolerance, supporting SFRP5 as a negative regulator of glucose metabolism. Acco...
متن کاملCirculating adipocyte fatty acid-binding protein levels are independently associated with heart failure.
A-FABP (adipocyte fatty acid-binding protein), one of the most abundant proteins in adipocytes, plays a key role in obesity-related insulin resistance, inflammation and atherosclerosis in animals. In the present study, we sought to investigate the association of A-FABP with HF (heart failure) in Chinese subjects. Serum A-FABP levels were measured in 252 HF patients and 261 age-, gender- and BMI...
متن کاملSecreted Frizzled Related Protein 3 in Chronic Heart Failure: Analysis from the Controlled Rosuvastatin Multinational Trial in Heart Failure (CORONA)
BACKGROUND We have previously demonstrated an association between increased sFRP3 expression and adverse outcome in a population of HF irrespective of cause and left ventricular ejection fraction. In this study we evaluated the prognostic value of sFRP3 in older patients with chronic systolic HF of ischemic origin. METHODS We evaluated sFRP3, by tertiles, as a risk factor for the primary endp...
